Diversité génétique de six populations de Heterobranchus longifilis de Côte d’Ivoire

1. RÉSUMÉ
La structuration génétique de Heterobranchus longifilis de côte d’ivoire a été étudiée à travers six populations. L’objectif de cette étude est d’identifier une ou des souches d’élevage de Heterobranchus. longifilis en Côte d’Ivoire, en vue de leur valorisation et vulgarisation. Ainsi, 116 échantillons provenant de 4 bassins versants (Agneby, Bandama, Cavally et Sassandra), ont été génotypes à l’aide de 6 marqueurs microsatellites isolés de Clarias gariepinus (Cga01, Cga03, Cga05, Cga06, Cga09 et Cga10). Une grande variabilité génétique intra population a été observée chez les différentes populations de Heterobranchus longifilis étudiées avec une diversité génétique élevée (HS=0,454) et un nombre moyen d’allèles observés de 8 ± 5,90. Sous l’hypothèse de Hardy-Weinberg, toutes les populations de Heterobranchus longifilis ont montré un déficit significatif à la panmixie, à l’exception de celle du Cavally. Les valeurs de FST (0,037 ; 0,078 ; 0,095 et 0,096) ont montré une différenciation génétique modérée entre les populations de Agneby nord, Agneby sud, Bandama et Cavally. Cependant, une forte différentiation génétique a été révélée entre les populations du Sassandra (nord et sud) et celles des autres (Agneby nord, Agneby sud, Bandama et Cavally). Les valeurs des FST se situent entre 0,16 et 0,27. Les regroupements des populations réalisés à partir de la matrice des distances génétiques de Cavalli-Sforza &
Edwards ont révélé une faible structuration génétique. Cependant, ces résultats indiquent l’existence de deux groupes bien distincts ; l’un composé de Agneby nord, Agneby sud, Bandama et Cavally et l’autre de Sassandra Nord et Sud.
SUMMARY
The genetic structuring of Heterobranchus longifilis from Côte d’Ivoire was studied across six population. The aim of this study is to identify one or more breeding strains of Heterobranchus. longifilis in Côte d'Ivoire, with a view to their promotion and popularization. Thus, 116 samples from 4 main watersheds of the hydrographic network (Agneby,Bandama, Cavally and Sassandra) , were genotyped using 6 microsatellite markers isolated from Clarias. gariepinus (Cga01, Cga03, Cga05, Cga06, Cga09 and Cga10). A large intra-strain genetic variability was observed in the different strains of Heterobranchus

longifilis, with high genetic diversity (HS = 0.454). Moreover, the average number of observed alleles was 8 ± 5.90. Under the Hardy-Weinberg hypothesis, all strains of Heterobranchus longifilis showed a significant panmixie deficiency, with the exception of Cavally. Otherwise, the FST values 0.037; 0.078; 0.095 and 0.096 showed moderate genetic differentiation between subpopulations of Agneby (North and South), Bandama and Cavally, repectively. However, the results of this study revealed a strong genetic differentiation between Sassandra populations (north and south) and Agneby populations (north and south), Bandama and Cavally with FSTvalues of 0.16; 0.19; 0.23; 0.24 and 0.27, respectively. The analysis of genetic structuration of Heterobranchus longifilis strains using the Cavalli-Sforza & Edwards genetic distance matrice, revealed weak genetic structuration.
However, the results showed that theses strains can be separated into two distinct groups:
one consisting of Agneby north, Agneby south, Bandama and Cavally and the other of Sassandra north and south.
