( 1 9 6 8 ) . F i l t r a t i o n s t u d i e s s u p p o r t t h e s e o b s e r v a t i o n s .

The v i r u s p a r t i c l e s ( B e r g o l d , 1962) a r e s p h e r i c a l , and show a l i m i t i n g membrane ( ? e n v e l o p e ) . He d e s c r i b e s and i l l u s t r a t e s t h e p a r t i c l e s i n a s t r o c y t e s o f mouse b r a i n a s a l w a y s a c c u m u l a t e d w i t h i n c i s t e r n s and d u c t s o f t h e e n d o c y t o p l a s m i c r e t i c u l u m . I n s a l i v a r y g l a n d c e l l s o f Aedes a e g y p t i he d e s c r i b e s p a r t i c l e s o c c u r r i n g f r e e i n t h e c y t o p l a s m and w i t h i n c i s t e r n s p r o b a b l y b e l o n g i n g t o t h e e n d o c y t o p l a s m i c r e t i c u l u m . O t h e r g r o u p B a r b o v i r u s e s h a v e been d e s c r i b e d a s b u d d i n g a l m o s t e x c l u s i v e l y upon p r o l i f e r a t i n g i n t r a c y t o - p l a s m i c membranes w i t h a c c u m u l a t i o n o f e n v e l o p e d v i r u s p a r t i c l e s w i t h i n d i s t e n d e d c i s t e r n a e (Murphy, 1968) and Murphy p r e s e n t s a s i m p l i f i e d c o m p a r a t i v e schema, w i t h g r o u p A a r b o v i r u s e s c h a r a c t e r i s t i c a l l y s h o w i n g an a r r a y o f n u c l e o i d s , g r o u p B a s above, and w i t h Bunyamwera

Y F v i r u s i s n o t a p p r e c i a b l y i n a c t i v a t e d by non-immune s e r a o f a number o f v e r t e b r a t e s p e c i e s (man, r a b b i t , mouse, monkeys o f many s p e c i e s , h a m s t e r s and g u i n e a - p i g s , h o r s e ) . The s i t u a t i o n v i s - a - v i s s e r a o f r u m i n a n t s i s n o t so c l e a r and i t i s p o s s i b l e t h a t s e r a o f some s p e c i e s , o r i n d i v i d u a l s may c o n t a i n a v i r u s i n a c t i v a t i n g s u b s t a n c e . The v i r u s w i t h s t a n d s d e s i c c a t i o n w e l l .

A number o f l i p i d s o l v e n t s and l i p i d e m u l s i f y i n g a g e n t s , s u c h a s sodium d e s o x y c h o l a t e , c h l o r o f o r m and e t h e r r a p i d l y i n a c t i v a t e t h e v i r u s , p o s s i b l y by s p l i t t i n g a l i p i d - c o n t a i n i n g e n v e l o p e , p e r m i t t i n g r i b o n u c l e a s e s t o a c t upon t h e i n f e c t i o u s v i r u s RNA.

Cheng (1958) has shown t h a t Y F , i n common w i t h o t h e r g r o u p B a r b o v i r u s e s he t e s t e d i s r a p i d l y i n a c t i v a t e d by p r o t e a s e s ( t r y p s i n , c h y m o t r y p s i n and p a p a i n ) , w i t h d e s t r u c t i o n a l s o o f h a e m a g g l u t i n i n s , where as t h e g r o u p A a r b o v i r u s e s r e m a i n u n a f f e c t e d . He p o i n t s out t h a t t h i s a c t i v i t y o f t h e enzymes i s a t r u e e n z y m a t i c a c t i v i t y , a p p a r e n t l y d i r e c t e d a g a i n s t t h e v i r u s p a r t i c l e s , r a t h e r t h a n an a c t i v i t y d i r e c t e d a g a i n s t o t h e r components o f t h e mouse b r a i n p r e p a r a t i o n s he u s e d , r e s u l t i n g i n a s e c o n d a r y v i r u s i n a c t i v i n g m o i e t y . T a k e h a r a e t a l . ( 1 9 6 1 ) showed t h a t t r y p s i n and a l s o p a n c r e a t i c d i p a s e i n a c t i v a t e d p a r t i a l l y p u r i f i e d Y F v i r u s .

They a l s o d e m o n s t r a t e d an i n a c t i v a t i o n o f WEE by t h e s e enzymes. The r e s u l t s o f t h e s e two r e p o r t s a r e n o t c o m p a r a b l e s i n c e d i f f e r e n t v i r u s p u r i f i c a t i o n m e a s u r e s w e r e e m p l o y e d , and e x p o s u r e t o enzyme p r e p a r a t i o n s was c o n s i d e r a b l y l o n g e r i n t h e T a k e h a r a e x p e r i m e n t s .

N i e l s e n e t a l . (1961) e x t r a c t e d i n f e c t e d baby mouse b r a i n w i t h p h e n o l , and showed t h a t t h e r e s u l t i n g m a t e r i a l , w i t h a U-V a b s o r p t i o n s p e c t r u m c h a r a c t e r i s t i c o f RNA had a b o u t 0.03%

o f i n f e c t i v i t y o f t h e o r i g i n a l p r e p a r a t i o n , and t h a t t h i s m a t e r i a l was r e a d i l y i n a c t i v a t e d by RNA a s e b u t n o t by DNA a s e . They a l s o showed t h a t t h i s m a t e r i a l was i n a c t i v a t e d by t r y p s i n .

T h e r e a r e no d e f i n i t i v e r e p o r t s o f t h e c h e m i c a l c o m p o s i t i o n o f Y F v i r u s . D e t e r m i n a t i o n s o f Ada e t a l . ( 1 9 6 2 ) f o r M u r r a y V a l l e y E n c e p h a l i t i s v i r u s o f 1 1 % l i p i d s , and RNA c o n t e n t

7.8%, w i t h a d e n y l i c a c i d 25.5, g u a n y l i c a c i d 27.5, c y t i d i l i c a c i d 21.5 and u r a d i l i c a c i d 25.5 may be r e l e v a n t . S t e l l a r e t a l , ( 1 9 6 6 ) r e p o r t f o r dengue 2 v i r u s , an RNA c o n t e n t o f 7%, and n u c l e o t i d e s i n t h e 45S ( i n f e c t i v e ) f r a c t i o n o f a d e n i n e 30.6, g u a n i d i n e 26,4, c y t i d i n e 21,3 and u r i d i n e 21,6.

Mussgay ( 1 9 6 4 ) p r e s e n t s a u s e f u l s y n o p s i s o f t h e g r o w t h c y c l e o f a r b o v i r u s i n v e r t e b r a t e and a r t h r o p o d c e l l s , w i t h i n f o r m a t i o n on t h e r a n g e o f v e r t e b r a t e and a r t h r o p o d c e l l s i n w h i c h Y F v i r u s c a n be grown. B o r r o w i n g from dengue 2 s t u d i e s , S t e v e n s e t a l . (1962) h a v e shown t h a t r e l e a s e o f i n f e c t i o n s dengue 2 v i r u s by KB c e l l s o c c u r s a f t e r an a v e r a g e l a t e n t p e r i o d o f 12 h o u r s and i n f u r t h e r s t u d i e s o f v i r u s k i n e t i c s S t e l l a r e t a l . ( 1 9 6 6 ) c o n c l u d e t h a t s y n t h e s i s o f e s s e n t i a l RNA b e g i n s s i x h o u r s a f t e r i n f e c t i o n o f c e l l s and p r e c e d e d r e l e a s e o f m a t u r e v i r u s by s i x t o s e v e n h o u r s . They d e s c r i b e a 45S-RNA f r a c t i o n , c o n t a i n i n g t h e i n f e c t i o u s ( s i n g l e s t r a n d e d ) v i r u s and a 20S-RNA f r a c t i o n c o n t a i n i n g d o u b l e s t r a n d e d m a t e r i a l .
