Resume : Apres une i n j e c t i o n de GaC13, on trouve du g a l l i u m dans l a chromatine e t l e s chromosomes, dans l e n u c l @ o l e e t dans l e s lysosomes.
A b s t r a c t : A f t e r an i n j e c t i o n o f GaClg, g a l l i u m i s found i n chromatin and chromosomes, i n n u c l e o l i and i n lysosomes.
Since g a l l i u m shows an a f f i n i t y f o r tumour t i s s u e , the r a d i o n u c l i d e 6 7 ~ a i s used as a d i a g n o s t i c agent f o r t h e d e t e c t i o n o f cancers by s c i n t i s c a n n i n g techniques. I t has been c l e a r l y shown t h a t 6 7 ~ a uptake by tumour c e l l s i s p r o p o r t i o n a l t o t h e m i t o t i c index (1). T h i s prompted our study on i n s e c t t e s t i s , together w i t h t h e f a c t t h a t we a r e c u r r e n t l y studying t h e a c t i o n o f chemotherapeutic agents on t h i s very same mate- r i a l .
I n t h i s EELS study, g a l l i u m was found i n chromatin, chromosomes, n u c l e o l i and lysoso- mes. Judging by t h e amount o f s t a i n a b i l i t y o f t h e c e l l u l a r o r g a n e l l e s i n m a t e r i a l n o t t r e a t e d w i t h osmium, g a l l i u m seems t o be a l s o present -but i n t o o small q u a n t i t y t o be detected- i n mitochondria and i n membranes, as w i t h indium. Not o n l y does t h e a c t i o n o f g a l l i u m resemble t h a t o f indium b u t t h e d i s t r i b u t i o n o f these two t r a n s i t i o n metals i s s i m i l a r . The house c r i c k e t s w e r e i n j e c t e d i n t r a - a b d o m i n a l l y w i t h 10 p1 o f one o f f o u r d i f f e r e n t aqueous s o l u t i o n s of GaCl the concentrations o f which were M (17.6 vg per i n - sect), 2 x 10-2 M, 4 x 10'3
M
and 10-1 M. The treatment f o r each dose l a s t e d 1 h, 2 h, 3 h, 4 h, 2 days, 3 days o r 7 days and 1 month f o r 2 x 10-2 M. The t e s t e s were r a p i d - l y dissected, f i x e d i n 2.5 % b u f f e r e d glutaraldehyde and p o s f i x e d i n 2 % b u f f e r e d OsO4. The m a t e r i a l was embedded i n Epon and the sections were 200 nm t h i c k . They were s t a i n e d w i t h uranyl acetate o n l y .The en.ergy l o s s spectra, corresponding t o s e l e c t e d areas 100 nm i n diameter, were ob- t a i n e d a t 1 MV using an R - f i l t e r and analysed w i t h t h e Tracor system connected t o t h e Toulouse 1.2 MV microscope. The Lp3 peak o f i r o n was found a t 730 eV and t h a t o f g a l - l i u m a t 1140 eV.
