2.2.2. Relation entre l’Intervalle de Tolérance et Erreur Totale
Suite à cette définition de l’intervalle de tolérance, on peut établir une relation entre celui‐ci et l’erreur totale. En effet, cet intervalle est toujours construit sur la base de la moyenne et l'écart‐type d'un échantillon d'un produit, par exemple. S’il est exprimé par rapport à la cible de la production d'un médicament ou par rapport à la valeur réelle des analytes dans un échantillon, l'intervalle de tolérance combine donc simultanément les erreurs aléatoires et systématiques d'une procédure analytique. Dans ce cas, la valeur moyenne estimée est remplacée par le biais estimé de la méthode d'analyse ou du processus de production. C'est donc un moyen statistique commode d'exprimer l’erreur totale [30, 62].
Les intervalles de tolérance peuvent être extrêmement utiles si on veut comparer les performances réelles d'un processus de production ou des méthodes d'analyse. En effet, pour les produits au moins, les spécifications sont établies afin d'évaluer leur qualité et leur donner l'autorisation d'être commercialisés. De l'examen d'un échantillon des produits, il est possible pour les producteurs de construire des intervalles statistiques de tolérance qui peuvent être comparés aux spécifications du produit et donc décider de sa libération sur le marché.
La même méthodologie peut être appliquée pour évaluer la validité des méthodes analytiques.
Hoffman et al., (2005 et 2007) [33, 62]ont utilisé un intervalle de tolérance de type « β – content » pour évaluer la validité des méthodes bio‐analytiques, alors que, une commission SFSTP [30] a proposé d'utiliser l'intervalle de tolérance « β – expectation » pour la validation des méthodes d'analyse. Brown et al. [69] ont proposé une méthodologie basée sur des intervalles de tolérance pour évaluer la bioéquivalence individuelle de deux formulations pharmaceutiques. Les intervalles de tolérance ont également été utilisés pour les tests de contrôle de qualité de l'uniformité de teneur d'un lot avant la libération du lot sur le marché [70‐73]. Les intervalles de tolérance pourraient également être utiles pour estimer la limite thérapeutique d'un médicament en dose études de recherche en veillant à ne pas tomber dans le niveau de toxicité ou d'inefficacité [79], ou en évaluant la sécurité des essais cliniques [80]
