propolis
L’éthanol est bien connu comme un agent ulcérogène, il induit des dégâts intenses aux niveaux
du tissu gastrique via des mécanismes multifactoriels, qui a été étudier par plusieurs auteurs
(Rtibi et al., 2015; Abebaw et al., 2017). Les polyphénols de la propolis possèdent un pouvoir
antioxydant important et bien décri dans plusieurs études (Teles et al., 2015).
Sur la base de nos résultats, nous pouvons indiquer les mécanismes par lesquels l’éthanol
intervient dans l’induction du stress oxydant et des dommages tissulaires gastriques d’une part,
et d’autre part celui par lequel les polyphénols de la propolis interviennent dans la lutte contre
ces dommages gastriques. L’éthanol administré par voie orale (1ml), provoque la production des
espèces réactifs de l’oxygène et mènent de ce fait à la consommation des molécules de défense
antioxydant comme le GSH, à l’augmentation de la peroxydation lipidique avec libération
massive du MDA. Ainsi qu’à la perturbation de la fonction enzymatique anti-oxydante (CAT,
SOD, GPx et GST) qui agit comme une importante défense cellulaire contre ces radicaux au
niveau de tissu gastrique, ce qui conduit au déséquilibre de la balance pro-oxydant-antioxydant.
Il affecte également les taux des paramètres gastriques, il augmente le volume de jus gastrique,
l’acidité totale et libre, réduise le pH et la production du mucus. Cette réduction de mucus
favorise de plus la génération de ROS. Le changement des paramètres du stress oxydatif et
gastriques se traduit sur le plan macroscopique et histologique par des lésions tissulaires, ulcères,
hémorragies, œdème, inflammation, nécrose et infiltration des neutrophiles. Les mécanismes
d’action possible impliqués dans l’activité ulcérogène d’éthanol sont élucidés dans la figure 22
ci-dessous.

L’extrait éthanolique de la propolis a montré une forte activité anti-oxydante in vivo ainsi qu’un
effet protecteur puissant contre la toxicité gastrique de l’éthanol. Il contient des composés poly-
phénoliquesnécessaires pour piéger et neutraliser les radicaux libres et protéger les lipides
membranaire de la peroxydation, que ce soit par captation directe des métabolites et des espèces
réactifs, ou par renforcement des systèmes antioxydants enzymatiques et non enzymatiques, et
donc le maintien de l’équilibre entre les po-oxydants et les oxydants. Les propriétés
anti-oxydantes des polyphénols résultent de leur réactivité élevée comme donneurs d’hydrogène ou
d’électrons et de leur capacité à stabiliser l’électron non apparié, ce qui confère à la propolis un
rôle important comme puissant antioxydants et assure ainsi la protection des fonctions gastrique
des toxicités liées au stress oxydatif d’éthanol.

Notre étude vise à évaluer in vitro l’activité anti-radicalaire des polyphénols de la propolis
algérienne de Kaous. Quantitativement, l’étude phytochimique a permis de déterminer la
présence des polyphénols et des flavonoïdes en proportion élevée dans l’extrait ethanolique de la
propolis.
Le potentiel anti radicalaire de l’extrait éthanolique de la propolis a été déterminé vis-à-vis le
radical libre DPPH° et le peroxyde d’hydrogène H
2O
2dont les résultats montrent que l’extrait
possède une bonne activité concentration-dépendante, due essentiellement à la présence des
polyphénols et des flavonoïdes.
La deuxième partie in vivo est consacrée à l’évaluation de l’effet gastro-protecteur et
anti-ulcérogène des polyphénols de la propolis contre l’ulcère induit par l’éthanol, notre étude est
concentrée sur l’évaluation de l’influence de l’administration d’éthanol par voie orale sur les
paramètres gastriques et certains marqueurs du stress oxydant. Nos résultats montrent que
l’éthanol est hautement toxique au niveaux de l’estomac via l’induction d’un état de stress
oxydatif par la formation excessive des radicaux libres induisant la péroxydation lipidique et un
déficit accru des systèmes antioxydants enzymatiques et non enzymatiques. Il affecte également
les taux des paramètres gastriques (volume, pH, acidité totale du suc et quantité de mucus
gastriques). De même, sur le plan macroscopique et histologique l’administration d’éthanol
conduit à l’apparition des altérations tissulaires sévères au niveau gastrique (ulcère, hémorragie,
nécrose, inflammation, œdème et infiltration des neutrophiles). Afin de lutter contre ces toxicités
et les réduire au maximum, nous avons procédé au traitement préventif naturel par la propolis,
qui a permis d’avoir un effet protecteur contre la toxicité gastrique d’éthanol par le renforcement
des systèmes antioxydants en augmentant les taux du glutathion, l’activité des enzymes
antioxydants (GPx, SOD, CAT et GST) et la réduction des niveaux du MDA, ce qui assure
l’équilibre de la balance prooxydant –antioxydant. De plus, la propolis assure cette protection
par le maintien et la conservation des taux des paramètres gastriques aux valeurs normales. Les
coupes histologiques réalisées sur des fragments des estomacs, permettent de confirmer l’effet
cytoprotecteur de la propolis par la réduction des dommages tissulaires.
Les résultats obtenus ont permis de mettre en évidence l’action protectrice de la propolis et sa
capacité à moduler les effets toxiques causés par l’éthanol.
Cette recherche nécessite d’autres études approfondies pour mieux se concentrer sur les effets
révélés. Il serait souhaitable de développer cette étude en mesurant d’autres paramètres de statut

antioxydant (GR, GSSG, vitamine C…) et des paramètres gastriques (histamine, pepsine,
gastrine et prostaglandines). Des études à l’échelle moléculaire sont aussi nécessaires pour
déterminer d’une part les composés actifs de la propolis, et d’autre part le mécanisme
moléculaire par lequel ces composés accomplissent leur rôle.
