VI. Démonstration de la pléiotropie du gène SOCS2 par les études génétiques

Dans le chapitre bibliographique (Chapitre I.), nous avons vu dans quelle mesure la protéine SOCS-2 avait un effet pléiotrope, c’est-à-dire jouant un rôle sur de nombreuses fonctions biologiques. Ceci est dû à sa nature de régulatrice de la grande voie de transmission de signal cellulaire qu’est la voie JAK/STAT. Nous avons ensuite affirmé l’association de la mutation R96C du gène SOCS2 à différents caractères tels que la santé mammaire, la production laitière, la croissance, la reproduction, etc. (Chapitre III.), validée par ailleurs grâce à l’étude prospective sur la prise en compte de cette mutation dans les modèles d’évaluation génomique (Chapitre II.). Ces associations ont toujours été démontrées de manière indépendante les unes des autres dans des modèles uni-caractère.

Bien que non développée dans ce manuscrit, une approche multi-caractère a été appliquée au cours de cette thèse : la méthode CLIP test (David et al., 2013), dont les résultats ont été présentés sous forme d’un poster associé à un papier court au congrès mondial de génétique animale appliquée aux animaux d’élevage (WCGALP) en février 2018 (Nouvelle-Zélande) (Annexe 22). Cette approche a confirmé que la région de SOCS2 était bien pléiotrope et que les différents marqueurs de cette région n’étaient pas associés de manière différente aux différents caractères (effets corrélés). Les caractères testés étaient la production laitière, le SCS et la bactériologie par qPCR pour des brebis en première lactation, et le Poids Âge Type à 250 jours des mâles des stations de testage. De nombreuses autres approches sont développées aujourd’hui pour mettre en évidence la pléiotropie de la détermination génétique des caractères (Hackinger & Zeggini, 2017). Il serait intéressant de tester certaines de ces méthodes et notamment celles, appelées randomisation mendélienne, permettant de déterminer si la corrélation entre deux caractères est due à un lien causal (i.e. le caractère 1 est un facteur de risque causal pour le caractère 2) ou à des facteurs confusionnels comme l'exposition environnementale. En effet, à ce jour, concernant l’association de la mutation à la production laitière par exemple, deux hypothèses sont émises : lien direct via la régulation de la prolactine, ou effet indirect via la régulation de l’hormone de croissance qui joue sur le gabarit des brebis et ainsi sur leur capacité de production.
