IS somatropin (rec human growth hormone)

Code 98/574

Project leader A. Bristow

Date of evaluation May 2009

Date of establishment 2000

Principle application(s) Primary: Physico-chemical assays for rec hGH

Secondary: Bioassays sing conventional specific activity Secondary: Immunoassays

Summary of Stability data obtained at establishment

6 month accelerated degradation study

No measurable loss of somatropin monomer at

temperatures below 45deg, therefore no predicted loss of activity.

Predicted loss of HPLC purity – 0.017%.year at -20

ECBS notes/follow-up recommendations at establishment

Committee noted that stability will continue to be monitored in real time

Summary of post establishment stability evaluation

At 11 years post-fill, ampouled materials (stored at -20) compared with Frozen baselines stored continuously at - 150deg.

No differences in:

- Monomer content (predicted biological activity) - RP HPLC purity profile

Recommendation for ECBS endorsement

ECBS is asked to note that real time stability studies have confirmed predictions at time of establishment

Proposed date of further

evaluation 2018 (20 yeas post fill) or on replacement, whichever is sooner
