INTRODUCTION
The culture of bumblebee colonies is performed for pollination of agricultural
crops
aswell
asfor scientific studies. Several attempts
weremade to improve seed production by augmenting bumblebees in the field : surface and underground
domiciles
were set outfor overwintered queens
orbumblebee colonies reared in
captivity
werebrought
tothe crops
atthe beginning of the flowering period (reviewed by H OLM , 1966 and A LFORD , 1975 ; Mncr·altLaNE et a[., 1983).
Bumblebees
werealso used in greenhouses for pollination in plant breeding and
even
in gauze cages for pollination of small multiplication units (MoRGNEIt, 1973).
In several laboratories bumblebees
arereared for scientific studies of social
behaviour, learning behaviour,
castedetermination, and others. For all of these purposes it is desirable
tobe able
toculture bumblebees throughout the year.
In temperate zones, bumblebees have
onegeneration per year. Only few
observations
werepublished that
asecond generation could exist in the field (M
EIDELL
, 1968 ; Ma!I.DE, 1981). Young queens which emerge during
summerand autumn synthesize the
reservesfor hibernation during the first days after
eclosion (A LFORD , 1969 ; P OUVR EAU, 1976). About the third day they leave the
nest
and
arethen ready
to mate.After copulation the queens become lazy, seek
suitable overwintering sites and
enterdiapause. The entering of diapause is
anendogenous queen
castecharacteristic independent of temperature and light.
Young queens left in the
nestin
acontrolled climate
roomsit apart from the
nestand do
notparticipate in household tasks. Investigations have shown that the
specific physiology of young queens is based upon
alow juvenile hormone titre in haemolymph (RbsELER, 1976 ; 1977). When newly emerged queens
aretreated with juvenile hormone, glycogen and lipids
are notaccumulated in the fat body, but oogenesis is induced and the queens
startegg-laying after
somedays. A CO!-narcosis has
asimilar effect (RöSELER and R6si 1.i R, 1984).
Queens overwintered in captivity
areoften difficult
toactivate. An increased temperature alone does
notbreak diapause. HoI.M (1965) has shown that queens of Bombus terre,stri.s hibernated in
agreenhouse under controlled conditions
emerged
over aperiod of
morethan
twomonths. Presumably
anendogenous
readiness for breaking diapause is necessary for queens
torespond
tochanges in
environmental factors. It is, however, possible
toactivate hibernated queens by C0
2
-narcosis
sothat they
startnest-building after
afew days.
Since C0 2 -narcosis
canbe easily performed, I have used this technique
to
inhibit diapause
aswell
as toactivate overwintered queens. By both methods it
is possible
to rearbumblebees year-round. In the present paper the technique is
described.
MATERIAL AND METHODS
Colonies of B. terrestris were reared in a controlled climate room at -!- 28° - + 30 °C and 60 % RH. In
spring,
queens were collected in the fieldprior
tostarting
a nest.Queens
with nestscan be
easily distinguished
from othersby
theirpollen collecting.
Varioustechniques
used forinitiating
a colony have beenpublished (reviewed by Ho!mt, 1966 ; AFFORD, 1975).
The two-compartment system described
by
HASSELROT(1960)
in whichsingle
queens were confined wasused in my
experiments.
The bees were fed withwater-honey
solution(50 : 50,
v :v)
andpollen
collectedby pollen-traps
fromhoneybees. Only
freshpollen immediately deep
frozen in smallplastic bags
afterbeing
collected in theevening
was used.At the climax of
colony development
new queens are reared. In B. terrestris there are nodifficulties in
mating.
When young queens had left the nestbox, they
wereplaced
in gauze cages(50
X 50 X 50cm)
with males from other colonies. Most of the queens mated betweenday
three and six after emergence.Very
seldom werematings
of queens older than 10days
observed.Young queens not used for immediate nest initiation were overwintered in a
refrigerator.
Twoto three
days
aftercopulation
queens wereplaced singly
inglass jars
lined with moist soft paper and stored at + 5 °C -+ 10 °C. Every
month the queens were transferred to otherjars
in casesof mold
development.
For
CO 2 -narcosis
queens were put in aglass jar
and treated with aCO.
stream untilthey
became immobilized.They
remained in the closedjar
for 30 min.RESULTS
Nest foundation without diapause
Young mated queens
wereconfined in gauze cages. One day after mating
queens
werenarcotized with CO! for 30 min. When they
werestill lazy the following day, the narcosis
wasrepeated. After CO!-treatment queens became very active and flew in the gauze cage. Two
tofour days following narcosis single
queens
wereconfined in
nestboxes. Sometimes they became broody
evenin the
gauze cage and started secreting
wax.Since colonies
weremostly available, single
queens
wereoften confined together with brood (old larvae
or afreshly pupated comb) and
some(2-5) newly emerged workers. This method facilitates
nestfoundation. Queens normally accepted the comb
onthe first day. Most of them started egg-laying within
oneweek (Table 1). Workers older than five days
wereremoved
toavoid competition for egg-laying, and
werereplaced by newly emerged
ones, if it seemed necessary, for feeding larvae
orfor fixing the comb. The colonies
started in this way normally developed except that the first brood occasionally
contained
somemales.
Activation of overwintered queens
The shortest duration of overwintering tested
wasthree weeks, the longest
six months. Shorter
orlonger periods may be possible. To activate the queens
a
soft C0 2 -stream
wasintroduced into the hibernation jars for 10
secin the evening. The jars
wereclosed and put in
arefrigerator
at+ 15 °C overnight. The
next
morning the queens
weretransferred
togauze cages kept
at roomtemperature.
Two
tothree hours later the queens
weretaken
outof the cages and stimulated
tofly in the
roomfor about 10 min. Thereafter they
werenarcotized with C0 2 for 30 min. In the afternoon the queens
wereagain stimulated
tofly. When they
were
still lazy the
nextday, the C0 2 -treatment
wasrepeated. Until
nestinitiation
the queens
wereallowed
tofly in the
roomtwice daily for 10 min. Three
tosix days after the last narcosis the queens
weretransferred
tothe climate
roomand confined in
nestboxes
asdescribed above. Using this
treatmentthe first eggs
were
also laid within
oneweek (Table 1).
DISCUSSION
The method given in this paper makes it possible
to rearbumblebees year- round. This is
notonly
truefor B. terrestris, but also for B. hypnorum and
B. pratorum which
wehave occasionally cultured in captivity. Using this technique
research is not limited
tothe
summerand, moreover, colonies
areavailable for
pollination during spring when only queens
orfew workers
arepresent in the field.
A substantial prerequisite for year-round rearing, however, is
tostore
asufficient
amount
of pollen.
