Syringe guide for instrumental insemination
apparatus of queen bees (Apis mellifera L)

Summary —
Thesyringe guide
consists of along
thick wire rod fixed to thesyringe
holder. This rodallows an inseminator to make all
syringe-tip
movementsby moving
theguide
with his left hand. Theright
hand is then free to insert thesyringe-tip
and semen into thevagina,
without the inconvenience ofremoving
the hand from thesyringe.
Theguide
enables queens to be inseminated morequickly
and with less discomfort. Construction of the
guide
issimple
and economical.Apis

INTRODUCTIONInstrumental insemination is now used on a
large
scale for mass insemination of queen bees. One inseminator inseminatesscores of queens per
day.
It is thereforeimportant
toimprove
theapparatus
insuch a way as to make insemination easy,
quick
and with a minimum of discomfort.Of course, the collection of semen from the drones is the most
time-consuming procedure.
The
procedure
of introduction of the sy-ringe tip
into thequeen’s vaginal
orificecan be
improved
upon.Originally,
Mack-ensen and Roberts
(1948)
as well as Laid-law
(1944, 1977)
used thevaginal probe
to lower the
vaginal
valve-fold. Thetip
was then introduced behind that
probe.
However to make insemination faster and
easier, professional
inseminators have discontinuedusing
the valve-foldprobe.
Introduction of the
syringe-tip
into thequeen’s genital
tractrequires
veryprecise
movements. In Mackensen’s
apparatus,
thetip
was loweredby pushing
the sy-ringe
down. Inclination of thesyringe (dor-
so-ventral movements in relation to the
queen)
was alsoperformed directly by
hand. The transversal movements
(right
and left in relation to the
queen)
were madeby revolving
the entiresyringe block,
with thesyringe
in theholder,
around the vertical
support.
Thistype
ofapparatus
is still usedby nearly
everyoneperforming
instrumental insemination of bees in the USA(Harbo, 1989).
Due tothe lever
principle,
small movementsmade
by
hand at the level of thesyringe
block holder result in
large
movements at thetip
of the needle. These hand-directedmovements made it difficult for the
inexpe-
rienced to introduce the
tip
into thegenital
orifice.
Vesely (1966)
therefore added a screwadjustement
to thesyringe
holder.By turning
a screw, thesyringe
inclinationwas
altered,
and thetip
of the needle was moveddorso-ventrally.
But the transversal movements of thetip
still had to be carried outby hand, rotating
the block holder with thesyringe
around the verticalsupport.
This
procedure
wasimproved
uponby Schley (1983),
who constructed anotheradjustment,
which enables transversal movements of thesyringe
with thetip.
Swienty
in Denmark has constructed aprecise apparatus provided
with 3 micro-manipulators (Holm, 1986).
Eachmanipu-
lator moves one of the 2 hooks or the sy-
ringe.
The movements are veryprecise;
however,
the hook’spossibilities
are solimited that the
queen’s
abdomen cannotbe
opened by
them. Pointedforceps
haveto be used to open the abdomen. Dorso- ventral or vertical
syringe
movements canbe made with the
help
of themicromanipu-
lator. But transversal movements of the hooks or the
syringe
have to be madeby turning
additional screws. Different modifi- cations of theapparatus
for instrumental insemination have been described in the latest manual on instrumental insemination(Moritz, 1989).
All the above
improvements
have cer-tainly presented
some progress in the de-velopment
of theapparatus
for instrumen- tal insemination.Nevertheless,
all theseadjustements
did not resolve theproblems
connected with the movements and intro- duction of the
syringe tip
into thegenital
orifice of the queen. All
tip
movements aswell as the introduction of the
tip
into thegenital
orifice had to be directedby
thesame hand. This
required removing
theright
hand from thesyringe
or the micro-manipulator
to turn the screw discs for ad- ditionaladjustments,
and eitherlooking
away from the
microscope
orturning
thediscs without
looking.
Thesyringe
holderor the
micromanipulator
becamecompli- cated,
difficult to construct andexpensive
to
buy.
RESULTS
Construction of the
syringe guide
A
simple
and usefulsyringe guide
hasbeen constructed in Poland
(fig 1). It
al-lows all
tip
movements to be directed withone
hand,
while the other hand is used to introduce thesyringe tip
into thequeen’s genital
orifice. Theguide
consists of along
thick wire rod fixed to the
syringe
holder(fig 2).
Due to the leverprinciple, large
movements at the end of the
guide
resultin small
precise
movements at thetip
ofthe
syringe
needle.Before the
guide
can be fixed to the ap-paratus,
thesyringe
holder must be fixedpermanently
to the head of the bolt onwhich the holder rotates
vertically
in the sy-ringe
block(fig 3a).
The bolt should be alsolonger
than in the Mackensen appara- tus. It shouldprotrude
2 cm above the nutwhich holds the
spiral spring
between itand the block.
Next a wire = 26 cm
long
and 6 mmthick is selected. The wire can be > 6 mm
thick,
but should not be toothin,
because itmight
then be too flexible forprecise
move-ments. A transversal hole should be drilled
near one end of the rod
(fig 3t).
The bolt of the queen holderslips through
the hole in theguide,
and theguide
is fastened to the bolt with 2 check nuts(fig
3n).
The 2 nutshold the end of the rod between them. It is
possible
to work with theguide
fixed in this manner, but it is better to use a set screwwhich
keeps
theguide
fromslipping
on thebolt. The set screw
penetrates
theproxi-
mal end of theguide (fig 3s).
A hole isdrilled from the end of the rod
along
itsaxis to the hole for the bolt.
Procedure of queen insemination with the
help
of theguide
Insemination of the queen is facilitated both
by
the use of theguide,
andrejection
of the
valve-probe. First,
thetip
is filledwith semen, and the queen is
prepared
forinstrumental insemination as usual. All the
movements at the
tip
of the needle aremade with the aid of the
guide by moving
itwith the left hand. The entire
procedure
ofthe introduction of the
tip
into thequeen’s vagina
is madeby
theright
hand withoutremoving
it from thesyringe.
First,
thetip
of the needle ispositioned
near the dorsal
edge
of thevaginal
orifice.Next it is inserted = 0.5 mm into the
vagi-
na. Then the distal end of the
guide
is liftedwith the left hand. As a
result,
thetip
of theneedle is moved
ventrally. During
thatmovement the valve-fold is
pressed against
the ventral side of thevagina,
re-sulting
inopening
of the entrance to the median oviduct. Thetip
is then inserted = 1.5 mmdeeper,
and the semeninjected through
the median oviduct into both later- al oviducts.It is even
possible
to make the 2steps
of
tip
introduction in one continuous inser- tion. While theright
hand monitors the in- troduction of thesyringe tip
into thevagina,
the left hand ensures that at the same time the
tip
is moveddorso-ventrally
with the aid of theguide.
After the semen has beeninjected
into thequeen’s reproductive
or-gans, the
tip
is withdrawn and insemina- tion iscompleted.

pour l’insémination artificielle des reines d’abeilles. L’introduction de lapointe
dans le tractusgénital
de la reinenécessite des mouvements très fins de la
seringue.
Dansl’appareil
deMackensen,
l’inclinaison de la
seringue
était obtenue manuellement par desdéplacements
di-rects de celle-ci et les mouvements trans- versaux en faisant
pivoter
tout le bloc de laseringue
autour dusupport
vertical. Par leprinciple
dulevier,
depetits
mouvementsimprimés
manuellement au niveau du blocseringue produisaient
des mouvementsamples
à lapointe
de laseringue remplie
de sperme.
C’est
pourquoi Vesely (1966)
a construitun
support
deseringue
avecréglage
parvis, qui permet
de modifier l’inclinaison de laseringue. Schley (1983)
a construit unautre
support qui permet
les mouvementstransversaux de la
pointe.
Mais la mani-pulation
des vis dessupports
comme l’in-troduction même de la
pointe
doivent êtrefaites avec la même main. Il faut pour cela que la main droite lâche la
seringue
pen- dant l’insémination. Lesupport
devenait trèscomplexe
et coûteux.Un
guide
pour laseringue, simple
etpratique,
a été construit à l’Institutapicole
de l’université
d’agriculture
de Varsovie(fig
1
g).
Ceguide
consiste en unelongue tige métallique
fixée ausupport
de laseringue (fig
2h;
b = blocseringue).
Par leprincipe
du
levier,
delarges
mouvementsimprimés
à l’extrêmité le
guide produisent
despetits
mouvements
précis
à lapointe
de la serin- gue. Lesupport
de laseringue
doit resterconstamment relié à la tête de la vis à la-
quelle
leguide
est fixé(fig 3a).
Pendant l’insémination artificielle de la
reine,
tous les mouvements de l’extrémité de lapointe
sont exécutés avec leguide
en le
déplaçant
avec la maingauche.
Toutle travail d’introduction de la
pointe
dans levagin
de la reine est réalisé par la main droite sans avoir à lâcher laseringue.
Cecipermet
d’inséminer des reinesplus rapide-
ment et
plus
facilement. La construction dece
guide
estplus
facile et moins coûteuseque celle des autres
dispositifs.

gefüllten Spritze.
Das hatteSchwierigkeiten
bei derEinführung
derSpritze
in dieGenitalöffnung
derKönigin
zur
Folge.
Deshalb konstruierte
Vesely (1966)
ei-nen
Spritzenhalter
mitStellschraube,
durch welche die
Neigung
derSpritze
verändert werden konnte.Schley (1983)
konstruierte eine andere
Vorrichtung,
welche
Bewegungen
derSpritze
in trans-versaler
Richtung ermöglichten.
Aber so-wohl die
Betätigung
der Schrauben dieserVorrichtung
wie dieEinführung
derSpritze
selbst in die Scheide hatten mit derselben Hand zu
erfolgen.
Deshalb mußte manwährend des
Besamungsvorganges
denGriff der rechten Hand zur
Bedienung
derSchrauben am
Spritzenblock
wechseln.Der
Spritzenhalter
wurde sehrkompliziert
und teuer.
Am Bieneninstitut der Landwirtschaftli- chen Universität Warschau wurde eine ein- fache und nützliche
Leitvorrichtung
für dieSpritze
konstruiert(Abb 1 g).
Die Vorrich-tung
besteht aus einemlangen,
dickenMetallstab,
der an demSpritzenhalter
be-festigt
ist(Abb 2h;
b =Spritzenblock).
Ent-sprechend
denHebelgesetzen
führen aus-giebige Bewegungen
am Ende desLeitstabes zu
geringen,
feinenBewegun-
gen der
Spritzenspitze.
DerSpritzenhalter
muß dabei fest mit dem
Kopf
der klemmsckraube verbunden bleiben
(Abb 3a);
zurwelche der Laitstab
befestig
ist.Während der instrumentellen Besa- mung der
Königin
werden alleBewegun-
gen des
Spritzenendes
mit Hilfe des Leitstabes durchBetätigung
mit der linken Handgesteuert.
Dergesamte Vorgang
derEinführung
derSpritze
in die Scheide derKönigin erfolgt
mit der rechtenHand,
welche dabei
ständig
an derSpritze
ver-bleiben kann. Dadurch kann die Besa- mung der
Königinnen
rascher und be- quemerdurchgeführt
werden als mitjedem
anderen
Apparat.
Die Konstruktion desLeitstabes ist einfacher und
billiger
alsandere
Vorrichtungen.
