INTRODUCTION
Studies on
detection, anatomy
andcytogenetics
of intersexgoats
have been de- scribed in several breeds in relation to the absence of horns in males(Padeh
etal, 1965; Ilbery
andWilliams, 1967;
Cribiu andLehrm, 1986;
Ramadan andEl-Hassan, 1988;
Moreno-Millan andRodero, 1988).
Ingeneral,
the presence or absence of hornscan be attributed to the action of two alleles at an autosomal
locus,
withpolled (P) being
dominant to horned(p).
In
goats,
horned(pp)
orheterozygous
forpolled (Pp)
XX individuals are normal females. But all XXgoats
that arehomozygous
for thepolled
allele are intersexes.In
addition,
aproportion
of XYgoats
that arehomozygous
for thepolled
allele aresterile.
In the
present
paper, we describecomparative
studies on thecytogenetic
andanatomic
configuration
of 3 cases ofintersexuality
ingoats belonging
to theMurciano-Granadina breed.
MATERIALS AND METHODS
Three intersex animals were
analyzed cytogenetically by preparation
ofleukocyte
cultures from
peripheral blood, according
to the method of Moorhead et al(1960)
with several modifications.
R-banding (Di
Berardino etal, 1987)
andG-banding (Francke
andOllivier, 1978)
were carried out toidentify
the chromosomepairs.
Sex chromosomes were studied in these animals
by
electronmicroscopy according
to the method
of Goyanes
and Mendez(1981).
Atpostmortem,
bone marrow,gonad,
kidney
and skin weresampled
for tissue cultures andcytogenetic
examination. At the sametime,
thereproductive
tracts were dissected and examinedmacroscopically
and the
specimens
were fixed in Bouin’s fluid and the sections were stained withhematoxylin
and eosin forhistological
studies.RESULTS
Two of the animals
(60,XX
and60,XY)
werepolled
and one(60,XY)
washorned, however,
all 3 wereexternally
considered females.Mosaicism,
chimerism or chro-mosome aberrations were not observed in any of the animals
(fig la, b).
The sex chromosomes studied
by
electronmicroscopy
showed a metacentric mor-phology
in the Y chromosome and subtelocentricmorphology
in the Xchromosome,
with a small short arm
(fig 2a).
A summary of the
macroscopic findings
on thereproductive
tracts in theseanimals is
given
in table I. All casespossessed
similar externalreproductive
organs andpresented
ahypertrophic
clitorisprotruding
from apseudovagina (figs
2b and3a). Although
thedegree
of masculinization varied amongthem,
their externalgenitalia
wereessentially
of the femaletype.
No scrotum wasobserved, although
2 testes were detected in the
inguinal region
of the two60,XY
animals.Only
2animals,
the60,XX
and one of the60,XY,
were dissected forhistological
and anatomic
analysis.
In the male60,XY,
the testes hadepididymides
and itsdeferent ducts had an
interrupted segmental
form.Emerging
from eachepididymis
were 2 ducts
ending
in anaplastic
uterus. Seminal vesicles were also present(fig 3b).
The60,XX
animal had a unilateral scrotum associated with theright
testis.The left testis was intra-abdominal. Both testes were
atrophic
withhypoplastic epididymides.
Two tubesemerged
from theepididymides.
One ended as anampulla
in a uterus of 8.4 x 2.8 cm,
which,
in turn ended in onecylindrical
tube connectedto the urethra. Two organs, identified as seminal
glands,
were found on the distalwall of the uterus.
Histological
studies of the clitoris of the60,XX
animal showed apenis-like
structure with connective and cavernous tissue. The testes exhibited neither meiotic
activity
nor germ cells in the dissected60,XX
and60,XY
animals.DISCUSSION
According
to the resultsobtained,
the animals studied can be defined as’pseudo- hermaphrodites’.
The association betweenintersexuality
andpolledness
ingoats
is summarized in table II. Our results are inagreement
with those of Nicholas(1987),
except
for the60,XY
horned male described. The reason for this association and thegenetic
determination are notyet known,
inspite
of several modelshaving
beenproposed
toexplain
them(Muller
etal, 1979;
Kent etal, 1986; Fredga, 1988).
The
discovery
ofrepetitive
Bkm(banded
krait minorsatellite)
sequences, asso- ciated with the male-determinantregion
on the Y chromosome in severalspecies (human,
mouse andhorse,
inparticular) (Kent
etal, 1988),
has led to the elabo-ration of a
hypothesis
as to theimplication
of these sequences in chromosome ar-rangements (deletions
ortranslocations). Consequently,
the studies of these’pseu-
dohermaphrodites’
are ofgreat
theoreticalimportance,
because of thelight they
shed on the mechanisms of sexual
differentiation
