C. Les neurones à kisspeptine
2. Kp dans le cerveau humain
Sur des échantillons d’autopsie de femmes pré- et post-ménopausiques, des neurones à Kp ont été localisés dans la partie caudale du noyau de l’infundibulum hypothalamique (équivalent de l’ARN chez les rongeurs)(Rometo et al. 2007) et ce groupe de neurones s'étend dans la partie proximale de la tige infundibulaire. Les neurones à Kp co-expriment la NKB et la DYN qui sont également essentiels dans la reproduction humaine. En effet, des mutations inactivatrices sur les gènes codant pour la NKB et son récepteur (NK3R) entraînent un HHI congénital normosmique avec de faibles niveaux de LH et une absence de puberté, malgré des taux de FSH quasi normaux (Topaloglu et al. 2009)(Young et al. 2010). Le cerveau humain n’a pas l’équivalence anatomique de l’AVPV des rongeurs, mais dans la région rostrale de l’hypothalamus, des neurones à Kp sont localisés au niveau des noyaux ventraux périventriculaires (PeN) et dans les subdivisions parvocellulaires antérieures des noyaux paraventriculaires (PVN)(Hrabovszky et al. 2010). Comme chez les rongeurs, les neurones à Kp dans cette zone préoptique présentent un dimorphisme sexuel avec plus de neurones chez les jeunes femmes que chez les hommes (Herbison 2008)(Hrabovszky et al. 2010)(Hrabovszky 2014)(Figure 13). Les populations de neurones à Kp du noyau infundibulaire et de la zone préoptique projettent sur les neurones à GnRH dans l'hypothalamus médiobasal (Hrabovszky 2014)(Figure 14). Chez l'humain, l'ARNm de KISS1R est fortement exprimé dans différentes régions du cerveau (cervelet, cortex cérébral et tronc cérébral), dans l'hypophyse et dans le placenta (Muir et al. 2001).

Kisspeptin neurons also show other robust sex differences in that the
number of KP-IR cell bodies and the density of kisspeptin-IR fibers are
much higher in aged women (B) compared with men (A). The number of
preproNKB-IR cell bodies is also higher in women (F) compared with men
(E), but the difference is less dramatic than in case of kisspeptin-IR cell
bodies. No obvious sexual dimorphism exists in the regional density of NKB
fibers (E–H). For quantitative comparisons, see Figures 2–4 . Scale

EXPERIMENT 2. INCIDENCE OF KISSPEPTIN-IR AND PREPRONKB-IR
CELL BODIES IN THE Inf
Quantitative analysis of the labeled cell bodies (using the
maxi-mal number of labeled cell bodies per 0.25 mm2 counting frame
for each male and female individual) revealed the following
differences
In males, preproNKB-IR cell bodies showed a significantly
higher incidence compared with kisspeptin-IR cell bodies
(P = 0.0005). PreproNKB-IR neurons outnumbered
kisspeptin-IR neurons by 120% (Figures 1A,E and 3).
In females, the mean incidence of preproNKB-IR cell
bod-ies was only 23% higher than that of kisspeptin-IR perikarya.
This subtle difference was not statistically significant (P = 0.28;
Figures 1B,F and 3).

Kisspeptin-IR cell bodies showed a 170% higher mean density
(incidence of cell bodies per 0.25 mm2 counting frame) in females
compared with males. This robust sex difference was statistically
significant (P = 0.002; Figures 1A,B and 3).
PreproNKB-IR cell bodies showed a 51% higher mean
inci-dence in females vs. males. This numerical sex difference was much
less conspicuous than for kisspeptin-IR cell bodies, but statistically
significant (P = 0.006; Figures 1E,F and 3).
EXPERIMENT 3. REGIONAL DENSITY OF KISSPEPTIN-IR AND
PREPRONKB-IR FIBERS
The Inf of aged men exhibited a few kisspeptin-IR fibers only, in
contrast with dense fiber networks in the Inf of postmenopausal
women (Figures 1A–D). PreproNKB-IR axons did not seem to

which is common to all forms of mouse KP [27] and is 90% identical to the corresponding human sequence (YNWNSFGLRF). Although the single amino acid sub-stitution at the C-terminal KP sequence of the human re-sults in a relatively low cross-reactivity (1%) of the #566 rabbit antiserum with the human KP-10 peptide in radio-immunoassay [27] , this antibody was still suitable for im-munohistochemical detection of human KP with the highly sensitive ABC technique and
silver-gold-intensi-fied nickel-diaminobenzidine chromogen [10] . A second polyclonal antiserum (GQ2; a gift from Dr. S.R. Bloom; London, UK) we used was raised in sheep specifically against the full-length KP-54 sequence of humans. This antiserum reacts with human KP-54, KP-14, and KP-10 and shows virtually no cross-reactivity (<0.01%) with other related human RF amide peptides, including pro-lactin-releasing peptide, neuropeptide FF, neuropeptide AF, and RF amide-related peptides (RFRP1, RFRP2, and
