Hypothèse 3. Certains des facteurs de transcription de type MYB-R2R3 sont impliqués dans la formation de la paroi secondaire du xylème
chez les conifères.
Ici, l’objectif a été d’analyser le rôle potentiel de certains gènes MYB-
R2R3 dans la formation de la paroi secondaire en manipulant leur expression par
transformation stable chez l’épinette blanche. Mes travaux ont ciblé trois gènes MYBs différents isolés du pin (Pinus taeda) et surexprimés chez l’épinette, un système d’expression "quasi-homologue". Ces travaux ont été rendus possibles de part la mise en place de plateformes spécialisées en transcriptomique et en transgénèse dans le cadre d’un projet de recherche ARBOREA.
I-3-2-Présentation des travaux de thèse
Mes travaux de thèse ont consisté en l’identification de plusieurs gènes codant des facteurs de transcription de type MYB-R2R3 en lien avec la formation du bois chez les conifères et, la caractérisation fonctionnelle de trois d’entre eux en système quasi-homologue par transformation stable chez l’épinette blanche.
Je présente dans un premier temps l’identification et l’analyse de séquences de 13 MYB-R2R3 de Picea glauca et 5 de Pinus taeda ainsi que les niveaux d’ARN messagers des MYB de P. glauca en lien avec la formation du bois chez l’épinette blanche (Chapitre II). Je montre que trois de ces gènes sont préférentiellement exprimés dans le xylème en différenciation de tige et de racine
ainsi que dans le bois de compression après 76 heures d’inclinaison de la même façon que certains gènes de la voie de biosynthèse des phenylpropanoïdes. Puis, dans une seconde partie, j’aborde la caractérisation fonctionnelle de PtMYB1 et
PtMYB8 par surexpression constitutive dans de jeunes plantules d’épinettes
blanches in vitro (Chapitre III), et dans des jeunes arbres transférés en sol dans le cas de PtMYB1 (Chapitre IV). Cette étude suggère fortement l’implication de ces gènes dans la régulation de la formation de la paroi secondaire lignifiée. Elle permet aussi de proposer l’existence d’un réseau impliquant des intéractions entre différents MYB-R2R3. Finalement, je présente la caractérisation d’un nouveau groupe de MYB appartenant au sous-groupe 4 (selon Krantz et al., 1998), le sous- groupe 4-C, qui est particulier aux conifères de part sa structure et sa composition. Ce sous-groupe est représenté entre autres par PtMYB14 qui a été soumis à une analyse fonctionnelle (Chapitre V et VI). Ce travail a nécessité la caractérisation du promoteur de l’alcool cinnamylique dehydrogénase (CAD) de P. glauca dont j’ai analysé le profil d’expression préférentiel aux tissus vasculaires (Chapitre V et VI). Mes résultats impliquent PtMYB14 dans les mécanismes de réactions de défense aux stress biotiques et abiotiques chez l’épinette blanche.
