spectrométrie de masse (MS native, approches top-down, middle-up, bottom-up,

Antibody-drug-conjugates(ADCs)manufacturingleadstoamixtureofspecieswhichneedstobe char-acterizedduringdevelopmentandforfurtherqualitycontrol.Thecouplingofon-lineHICxRPLCto highresolutionmassspectrometrycanbeconsideredasaveryefficientanalyticalmethod,providing extensiveinformationonADCsample,withinareducedtimescale.Ourintentioninthisfirstpaperisto presenttheapproachusedtorationallyoptimizethenumerousconditionsthatcanaffectthequalityof the2D-separation.HICandRPLCconditionswerethereforeoptimizedtopreventsaltprecipitationdueto solventmixingandtoenhancesensitivity,whilelimitingthetotalanalysistime.Wedemonstratedthat addingsaltinthesamplesolventbeforeHICinjectionallowsasignificantpeakshapeimprovement.The gradientprofilewasalsocarefullyoptimizedinbothdimensions,leadingtoatwo-stepgradientinHIC andbracketedgradientinRPLC.Thisstudyshowsthaton-lineHICxRPLChyphenatedtohighresolution massspectrometryisausefulmethodtoobtainrapidandextensivestructuralinformationonthepeaks observedinthefirstHICdimension,therebyleading,inasinglesteprequiring75min,totheprecise determinationoftheaveragedrug-to-antibodyratio(DAR)byHICaswellastheknowledgeofthedrug loaddistributionforaparticularDAR.ThestructuralcharacterizationofADCfragmentsbyRPLC-QTOF willbediscussedinthesecondpartofthistwo-partseries.

1. Introduction
Monoclonalantibodies(mAbs)andtheirrelatedproductssuch as antibody-drug-conjugates (ADCs), bispecific antibodies, Fab fragments,Fc-fusionproteinsandradio-immunoconjugatesarethe fastestgrowingclassofhumantherapeutics[1].Evenif numer-ousmAbsandderivativeshavebeenapproved,thefirstgeneration mAbsoftensuffersfromalimitedefficacy.ADCtechnology com-binesthespecificityofamonoclonalantibody(mAb)togetherwith apotentcytotoxicdrugcovalentlyboundedviaalinkertothe anti-body,allowinganefficacyimprovement.Therearetwomaintypes

ofADCtechnologybasedeitheroncysteineorlysinelinked conju-gationsandcurrently−asathirdclass−theso-calledengineered ADCs(cysteineresiduesareengineeredonthesurfaceofthemAb) areoftenmentionedasapromisingtechnology.
