4 1. Introduction Human chorionic gonadotropin (hCG) and follicle-stimulating hormone (FSH) belong to a family of related hormones called gonadotropins [1,2]. They are heterodimers sharing the α-subunit structure non-covalently bound to a specific β-subunit resulting in differences in their physiological roles and signaling. The α-subunit has 2 N-glycosylation sites , whereas the β-subunit of hCG has 2 N- and 4 O- glycosylation and of FSH contains 2 N-glycosylation sites. Similar to other hormones such as luteinizing hormone, hCG and FSH are used as therapeutic agents in reproductive medicine. However, their glycosylation state has an impact on their stability, solubility, biological activity, potential toxicity, and clearance [3]. This is why their characterization is required throughout their lifecycle (discovery, preclinical, and clinical steps), production, storage, and delivery.

Pharmacodynamics of follicle stimulating hormone (FSH) in postmenopausal women during pulsed estrogen therapy: Evidence that FSH release and synthesis are controlled by distinct pathways

F SH IS A heterodimeric hormone secreted from the pitu- itary. It is composed of an ␣-subunit that is common to LH, TSH, and human chorionic gonadotropin and a ␤- subunit that is unique. Two carbohydrate chains are linked to both ␣- and ␤-subunits. Glycosylation of the subunits is involved in the biological activity of the hormones, gener- ating 20 circulating isoforms of FSH. Gonadotropin secretion is under the control of GnRH and gonadal peptides such as activins, inhibins, and gonadal steroids [progesterone and 17␤-estradiol (E2)] (1). FSH is considered to be one of the main hormonal markers of menopausal status (2), as levels of the hormone are markedly increased at menopause. One year after menopause, FSH levels are 10- to 15-fold higher than those during the follicular phase in women of repro- ductive age (3–5). Estrogen replacement therapy has been shown to produce a dose-related decrease in serum FSH levels in postmenopausal women (6) after oral E2 adminis- tration (7) as well as after E2 administration by nonoral routes, such as transdermal, sublingual, and intravaginal estrogen (8 –11). Although nyctemeral variations in FSH lev- els and during the menstrual cycle have been well charac- terized, specific data from postmenopausal women receiving hormone replacement therapy are limited.

Hair Follicle miRNAs: a Novel Biomarker for Primary Blast Induced-Mild Traumatic Brain Injury.

The whisker follicle gene expression profiling data were collected, validated and submitted by our group previously (Zhang et al., 2018) (GEO Accession ID: GSE80371). Briefly, the microarray was conducted using the Agilent SurePrint G3 Rat GE 8 x 60 K system (Agilent, ON, Canada). Samples were prepared with 200 ng total RNA using the Agilent Low RNA Input Fluorescent Amplification Kit (Agilent, ON, Canada). Raw expression data were processed and analyzed using JMP Genomics 6.0 software (SAS, NC, USA). Gene level statistics (differential expression or DE analysis) were calculated using One-way analysis of variance (ANOVA) after Quantile normalization and removal of control data in JMP Genomics.

19 patients had repeated unexplained embryo implantation failure (IF) [13] defined as an absence of pregnancy despite the transfer of 10 or more good quality embryos (fresh or frozen-thawed) over several cycles. All embryos had a fragmentation rate of less than 20% and were at least at the 4-cell stage by day 2 after IVF. The inclusion criteria also included a normal uterine morphology (excluding fibroma, polyps, thin endometrium, synechia, uterine malformation) after ultrasonography, hysterosalpingography and hysteroscopy, a normal karyotype, a normal hormonal reserve (FSH < 10 mIU/ml, antral follicle count > 9 follicles) and normal responses to the hormonal stimulation (with more than 8 oocytes retrieved per cycle). Normal endometrial trophicity was defined after ultrasonography by an endometrial thickness over 7 mm and an endometrial volume over 2.5 cm3 during luteal phase. In this group, the mean age was 31.7 years (from 28 to 35) and the mean BMI was 22.6 (from 19 to 29).

Use of different postmenopausal hormone therapies and risk of histology- and hormone receptor-defined invasive breast cancer.

In an earlier report we examined the relationship between different types of HT and breast cancer risk, considered as a single disease, in the French E3N cohort. 6 We found that the risk was significantly lower with CHTs containing progesterone or dydrogesterone rather than other progestagens.We also observed a significant increase in risk with unopposed estrogens. We now examine whether the associations of these four types of HTs with breast cancer risk vary across different types of carcinomas, characterized by histological type and hormone receptor status.

Résumé Au cours du développement, les organismes croissent de façon harmonieuse suivant un programme génétique intrinsèque et en adaptation avec les conditions environnementales. Chaque tissu atteint une taille cible qui est proportionnelle à la taille finale des autres organes et à celle de l’organisme. Des expériences de régénération tissulaire effectuées sur différents modèles animaux ont révélé que chaque organe possède un programme autonome de croissance. Ainsi, des mécanismes de coordination entre la croissance tissulaire et le programme de développement sont nécessaires afin d’assurer une régulation fine de l’allométrie avant le passage du stade juvénile au stade adulte fixant la taille finale du corps. Dilp8 (Drosophila Insulin Like Peptide 8) est une hormone analogue aux peptides de la famille des insulines/relaxines récemment identifiée au laboratoire. Elle est produite par les tissus dont la croissance est lésée, en régénération ou néoplasique. dilp8 inhibe la production d’hormone stéroïde (ecdysone) et retarde ainsi le passage à la forme adulte (métamorphose). Adiposity, hormone replacement therapy use and breast cancer risk by age and hormone receptor status: a large prospective cohort study.

ER+PR+ breast cancer among women younger than 49 years progressively turned into a direct relationship among women of more advanced age. Furthermore, we observed increases in the risks of both hormone receptor-negative and -positive tumors among women who used HRT at baseline, although the increase in risk was weaker for ER- PR- than for ER+PR+ tumors. The HRT-related increases in risks of both ER-PR- and ER+PR+ breast cancer were stronger among the leaner women, and this increase was independent of HRT type. Finally, with respect to measures of body fat distribution (waist and hip circumferences) no relationships with either receptor-negative or -positive breast cancer were observed after adjustment for BMI. 26 1) Introduction Comme cela a été mentionné précédemment, peu d’informations sont disponibles concernant le devenir à long terme des patients traités par hormone de croissance dans l’enfance (41,50). En raison des propriétés mitogéniques et prolifératives de l’hormone de croissance, une grande attention a été portée sur le risque de cancer après les traitements par hormone de croissance (44,51). Les résultats préliminaires portant sur la cohorte française de l’étude SAGhE ont montré une augmentation de la mortalité cardio- et plus spécifiquement cérébro-vasculaire après traitement par hormone de croissance (48). Plusieurs études ont également démontré que les patients acromégales avaient un excès de risque de mortalité en lien avec les maladies cardio-vasculaires et cérébro-vasculaire, indépendamment des autres facteurs de risque (52,53). Il est également décrit une augmentation de la prévalence des anévrysmes intra-crâniens chez les patients acromégales, en association avec l’excès de GH dans le sérum au diagnostic de l’acromégalie (54). Ces observations et les effets clairement démontrés de l’axe GH-IGF-I sur les vaisseaux sanguins (55,56) nous a amené à étudier la relation entre l’exposition au traitement par hormone de croissance dans l’enfance et le risque d’accident vasculaire cérébral (AVC) à l’âge adulte, sur une population alors âgée de moins de 40 ans.
