RESUME
Cette recherche a été menée dans I'optique de déterminer le potentiel oxydant des molécules anticancéreuses,
utilisées de nos
jours
en clinique, lors de leur contact direct avec les globules rouges (GRs) rr?vitro.
Un protocole expérimental a été réalisé afin de révéler l'action des anticancéreux sur les érythrocytes humains.La cytotoxicité a été évaluée par la détermination de l'hémolyse par la mesure de la concentration cellulaire ainsi que le dosage de l'hémoglobine (Hb) libérée. Notre étude a dévoilé que ces molécules provoquent une
diminution de la concentration cellulaire des GRs ainsi qu'une hémolyse cellulaire importante qui dépend de
I'agent anticancéreux utilisé.

stress oxydant survenusur
la
membrane(cytotoxicité membranaire) qui s'explique en partie par I'oxydation des acides gras polyinsaturés qui aboutit
à
la
formation
des malondialdéhydes(MDA)
engendréspar les
espèces réactives de I'oxygène (ERO) produites par les anticancéreux; ou bien parl'effet
oxydant direct de ces demiers. Ce phénomène, conduit ainsi à la détérioration de la membrane et à la libération des constituants cellulaires des GRs. Le test deI'Hb
arévélé une oxydation remarquable deI'Hb
par certains anticancéreux. La comparaison de ces résultats avecles bilans
hématologiques des patients aprèsles
cures chimiothérapiquesa
dévoilé
clairement que lesmolécules anticancéreuses ont un potentiel oxydant sur les GRs. Cela concorde avec nos résultats
in vitro
déterminés pour la première fois par le contact direct avec les GRs.

This research was carried out
with

have found that theoxidative
effect
of
the
anticancer molecules causedby
oxidative
stresson the
membrane (membranecytotoxicity) can be explained in part by the oxidation of the polyunsaturated fatly acid, which results in the
formation of
MDA
by the ERO produced by anticancer agents. Leads to its denaturation and to the releaseof
the cellular constituents of the red blood cells. To unmask all the actions of these molecules, we went a little further
by
attributing the hemoglobin test which consistsin
bringing these moleculesinto
contâctwith
thehemoglobin where a hemoglobin oxidation was revealed for certain molecules. These results were compared
with
hematological assessmentsof
patients after chemotherapy.All
the results obtained also clearly showthat the anticancer molecules have an oxidizing potential on the red blood cells revealed for the first time by
the direct contact with the erythrocytes in vitro.
