Figure legend S1: In addition to the normal profile of sphingolipids observed in the extract of [3H]inositol labeled cwh43∆ cells (Fig. 2C), we also analyzed by TLC the lipid extracts of cells labeled with [3H]inositol (I) or [3H]DHS (D) obtained in the experiment shown in Fig. 2A. As can be seen in the above Figure, the profiles of ceramides and IPCs, whether treated (+) with mild base (NaOH) or not (-) are the same in wt and cwh43∆ (∆) cells. [3H]inositol-labeled lipids of wt cells are included in order to allow the localization of IPCs and mannosyl-IPC (MIPC). It has to be noted that cells can degrade [3H]DHS to [3H]palmitate and that this label then is

incorporated into a multitude of glycerophospholipids. The lowest band in the region of apolar lipids represents free fatty acids, and this band strongly increases upon mild base treatment. The region of apolar lipids also contains ceramides. Data argues that the metabolism and the incorporation of [3H]DHS in cwh43∆ cells is normal.
