Root hairs, tubular-shaped tip-growing cells that arise from root epidermal cells known as trichoblasts, are present on roots of all major groups of vascular plants, indicating their long evolutionary history.
They are thought to increase the absorptive capacity of roots by increasing the surface area. Root hairs also play a significant role in the establishment of interactions between plants and nitrogen-fixing micro-organisms (e.g. Rhizobium, Frankia) and symbiotic mycorrhizal fungi (Peterson and Farquhar, 1996).
In order to isolate genes specifically expressed in root-hairs, differential screening was performed on a Vigna unguiculata root hair. cDNA library (Krause et al., 1994) using two pools of single-stranded cDNA probes for the hybridization: one was synthesized from mRNA.s extracted from roots, the other from root hairs. Thirty clones were isolated after screening of20,000 recombinant phages. Of these, one phage carried an insert of approximately 1.2 kb which cross-hybridized only with itself After sub-cloning, the DNA sequence was determined and compared with E11BL database entries. Significant sequence homology was found with the TED2 locus of Zinnia elegans (Demura and Fukuda, 1994).
Overall homology was 66% at the DNA level while the deduced amino acid sequence displayed 67%
identity and 81 % similarity. In addition, homology to different NADH:quinone oxidoreductases was detected. TED2 of Zinnia elegans currently has no defined in vivo function but is specially expressed in developing vascular systems and was thought to be a possible marker for very early stages of
development. It might be therefore, that VuTED2 is a marker gene for early developmental changes in root hairs during nodule formation. :
Southern blots of genomic DNA digested with various enzymes suggested that VuTED2 is a member of a low-copy gene family.
