La méthode que nous proposons a été mise au
point
pourséparer
et doser simultanément lespentoses,
leshexoses,
les di- et triholosides à l’étatlibre,
ou obtenusaprès hydrolyse, dans les
contenus de rumen.
L’échantillon à
analyser
estpréalablement lyophilisé ;
il subit une éthérificationqui
trans-forme les fonctions -
hydroxyles
desglucides
en - éthers 0triméthylsilylés qui
sont volatils.- ) C―OH ――――――――> ! C―0 ― Si(CH!
Ceux-ci sont
séparés
parchromatographie
enphase
gazeuse. L’utilisation d’un standard interne(Inositol) permet
d’évaluerquantitativement
les différentspics
obtenus.Les modifications que nous avons
apportées
aux méthodesclassiques
dechromatographie
en
phase
gazeuse desglucides
sont les suivantes :a)
Au cours de la réaction desilylation,
lesglucides
sont dissous non pas dansla pyridine (S
WEELEY
etal., 19 6 3 )
mais dans lediméthylformamide (E LLIS , 19 6 9 ) qui permet
d’obtenir desrésultats
quantitatifs plus précis,
surtoutlorsque
l’on veutséparer
lespentoses.
b)
Lesquantités
des réactifs desilylation (hexaméthyldisilazane
ettriméthylchlorosilane)
ont été déterminées de
façon
à doserquantitativement
lesglucides
même sil’échantillon n’est pas parfaitement anhydre.
c)
Laphase
stationnaire utilisée est une résine siliconediméthyl
nonpolaire (OV,)
dont latempérature
limite d’utilisation est élevée( 350° C),
cequi permet
l’élution des triholosides à320 0 C.
Cette méthode
permet
uneséparation complète
de laplupart
desglucides simples
que l’on rencontregénéralement
dans les milieuxbiologiques. Cependant,
certainsglucides
sont malséparés (arabinose-rhamnose
etfructose-mannose)
et il sera sans doutenécessaire,
pour résoudre ces pro-blèmes, d’utiliser
des colonnescapillaires.

SUMMARY
QUANTITATIVE
GAS CHROMATOGRAPHY OF MIXTURES. OF
OSES,
DI - AND TRIHOI,OSID!SThe method
proposed
in thepresent
paper has been elaborated with the view of a simulta- neousseparation
andquantitative determination
of the freepentoses, hexoses,
di- andtriholosides,
or those obtained after
hydrolysis,
in the rumencontents.
The
sample
to beanalysed
ispreviously lyophilized ;
itundergoes
an etherification which transforms thehydroxyle
functions ofcarbohydrates
into volatile 0trimethylsilylated
ethers.which were
separated by
gaschromatography.
The utilization of an internal standard(Inositol)
allowed a
quantitative
evaluation of the differentpeaks.
The classic methods of gas
chromatography
ofcarbohydrates
arechanged
as follows :a) During
thetrimethyl silylation,
thecarbohydrates
were not dissolved inpyridine (S WEELEY
et
al., 19 6 3 ),
but indimethylformamide (E LLIS , 19 6 9 )
which resulted in formation of hexa-methyldisiloxane giving
more accuratequantitative results,
inparticular
whenseparating pentoses.

The amounts ofsilylation reagents (hexamethyldisilazane
andtrimethylchlorosilane)
weredetermined so as to obtain a
quantitative dosage
of thecarbohydrates
even if thesample
was notperfectly anhydrous.
c)
Thestationary phase
used was a nonpolar dimethyl
silicone resin(OV 1 )
with ahigh
tempe- rature limit( 35 o o C) allowing
elution of the triholosides at320 0 C.
By
means of this method it ispossible
to realize acomplete separation
of most of the sim-ple carbohydrates generally present
inbiological
media.However,
somecarbohydrates
are not wellseparated (arabinose-rhamnose,
andfructose-mannose) and,
to resolve theseproblems,
it willprobably
