Localisation of phospholipases D1 and D2 in the Golgi membranes of cells specialised for secretion.

Phospholipid metabolism and lipid-modifying enzymes play a key role in vesicular transport in eukaryotic cells. As previously shown in some mammalian cell lines, we recently demonstrated that phospholipase D (PLD) is implicated in the transport of secretory proteins in the exocytic pathway of mammary epithelial cells (MEC). Based on the observation that ARF stimulates PLD1, it has been suggested that this isoenzyme regulates transport in the secretory pathway. However, since there is no selective inhibitor of PLD isoenzymes, an additional role for PLD2 in this process can not be excluded. In the present study, we focused on the subcellular distribution of PLDs in two cell types specialised for secretion; namely rat exocrine MEC and the neuroendocrine cell line PC12. Using immunofluorescence, we found that both PLD1 and PLD2 were exclusively present in the perinuclear area of these cells (except that PLD2 label was also observed in the nucleus of PC12 cells), and weakly overlapped with TGN38, a marker of the trans-Golgi network (TGN). In contrast, PLDs immunoreactivity remarkably co-localised with the Golgi membrane of PC12, as identified by the median Golgi marker mannosidase II (for unknown reason, fluorescence of mannosidase II was not observed in MEC). This was consistent with the observation that this pattern of fluorescence disappeared in PC12 cells treated with brefeldin A. Finally, as we previously observed for caseins in MEC, butanol treatment of PC12 cells reduced the transport of the secretory marker chromogranin B to the TGN by ≈30%. In light of these results, it will be interesting to elucidate which of the PLDs operate in the formation of transport vesicles from the endoplasmic reticulum, intra-Golgi transport vesicles and secretory vesicles from the TGN.
