In thé traditional medicines of Europe, thé u a t e r e x t r a c t of FilipenduJa uJ.varia flouers has been used as anti-inflammatory, analgésie and diuretic ( 1 ) . The a c t i v e principles of this plant are knoun to be salicylic acid d e r i v a t i v e s and f lavonoids. 2 ' . Spiraeoside is thé major and characteristic flavonoid of Filipendula ulmaria flouers and, for this reason, ue determined thé amount of it by HPTLC densitometry. We measu-red thé fluorescence of spiraeoside a f t e r d e r i v a t i z a t i o n by diphenyIboric acid-2-amino-ethylester ( 3 ) . The measurement was achieved by means of a TLC Scanner program-me d to uork in reflection-fluorescence at 360 nm (Mercury lamp; eut off filter 450nrrv. We respected thé follouing chromatographic procédure:

- L a y e r : HPTLC plates silicagel 60 Merck

- Mobile phase: Ethylacetate-Formic acid-Water: 6:1:1

- Standard solution ( 0 . 2 and 0.4 ul): 4 mg of spiraeoside SCR in 10 ml methanol - Sample solution ( 0 . 2 vil): 0 . 2 5 0 g of Filiper.ï^la flou/ers were e x t r a c t e d by 25 ml

methanol 60°C (2 hours). The solution uas e v a p o r a t e d and dissolved in 10 ml méthane!. A f t e r linéarisation, thé concentration of spiraeoside uas estimed by measurement of thé différent standards and samples mean areas. In our findings, thé mean content of spiraeoside uas 3.1 %.

The repeatability, reproducibility and thé good linearity uere confirmed by valida-tion of thé method.

In France and Belgium, leaf e x t r a c t from Fraxlnus excelsior are especially used as anti-inflammatory ( 1 ) . This property could be at least partially explained by thé ability of some coumarins to inhibit thé formation of leucotrienes in polymorphonu-clear leucocytes ( 2 ) . For this reason, ue determined thé amount of fraxin by HPTLC densitometry. Like other hydroxylated coumarin glucosides, fraxin shous a blue fluo-rescence under UV light (366 n m ) ; this property uas used for its quantitative estima-tion. The measurement uas achieved by means of a TLC Scanner, programmed to uork in reflection fluorescence at 350 nm (Mercury lamp; eut off filter: 450 n m ) . We respec-ted thé follouing chromatographic procédure:

- Layer: HPTLC plates RP 18 Silicagel 60 Merck uith concentrating zones - Mobile phase: Phosphoric acid 0.2 % - Acetonitrile (60:40)

- Standard solution: 5 mg of fraxin SCR uere dissolved in 100 ml MeOH-H20 (1:1) - Sample solution: 0 . 2 5 0 g of Fraxinus leaves uere e x t r a c t e d by 25 ml MeOH at 40°C

( 9 0 mins). The solution was evaporated and dissolved in 10 ml MeOH-H20 (1:1)

- Applications of 1 and 2 ul for thé standard and 2 jl for samples.

A f t e r linéarisation, thé concentration of f r a x i n uas estimated by measurement of différent standards and samples mean areas.

We are testing several commercial batches to establish thé mean content of f r a x i n in

Fraxinus leaves.
